Here we present MAGIC-seq (microfluidics-assisted grid chips for spatial transcriptome sequencing), which leverages carbodiimide chemistry, spatial combinatorial indexing, and continuous crisscrossed microfluidics designs, enabling sensitive and stable profiling of diverse tissue types with eightfold increase in throughput, minimal cost (~$0.11/mm2), and reduced batch effects. MAGIC-seq breaks conventional microfluidics limits by enhancing barcoding efficiency through an innovative splicing grid design, facilitating compatibility between capture area and resolution. This advancement allows for previously unattainable full coverage of postnatal mice (up to 21.6 mm × 21.6 mm and 202,500 spots) and comprehensive elucidation of cellular structures in a mouse brain at near single-cell resolution.
