Evolved pQGS sequencing in E. coli. Escherichia coli BW25113

The pQGS plasmid was propagated in E. coli BW25113 for 30 in vitro to investigate the stability of the IncQ. The pQGS plasmid carried a costly gfp gene driven by a constitutive strong promoter. During the evolution, the GFP expression was knocked down or knocked out. Individual colonies were Sanger sequenced for the gfp gene mutations, gfp deletions and plasmid structure variations.
