Long noncoding RNA MIR100HG has been implicated in various tumors; however, its role in immune regulation remains largely underexplored. In the present study, we demonstrate that MIR100HG exhibits a downregulated expression profile in human gingival tissues affected by periodontitis. Notably, our observations reveal that the knockdown of MIR100HG significantly mitigates periodontal inflammation in murine models, thereby supporting the critical role of MIR100HG in the pathogenesis of periodontitis. Cell-based experiments demonstrate that the knockdown of MIR100HG significantly suppresses the expression of inflammatory factors in cells, while concurrently inhibiting the activation of nuclear factor kappa B in human gingival fibroblasts . Furthermore, mechanistic studies elucidate that MIR100HG interacts with the RNA-binding protein quaking , which serves as a negative regulator of the NF-KB signaling pathway. Collectively, our findings illustrate that the silencing of MIR100HG exerts a pronounced anti-inflammatory effect by modulating the NF-KB signaling pathway in HGFs, thereby suggesting that it may serve as a potential therapeutic target for the treatment of periodontitis
