研究目的:通过对携带不同Notch1等位基因的内皮细胞进行RNA测序(RNA-Seq)分析,探究Notch1剂量变化所导致的转录差异。实验方法:采用荧光激活细胞分选术(Fluorescence-Activated Cell Sorting, FACS),我们针对发育过程中不同Notch1等位基因背景下的内皮细胞生成了高通量RNA测序数据。实验结果:Notch1剂量可改变部分内皮基因的表达模式。本次研究对胚胎发育第9.5天(e9.5)分离得到的内皮细胞开展了RNA测序,所用胚胎携带多种Notch1等位基因,包括N1+/+、N1+/-、N1+/vg、N112/vg及N112/-。
