Rat orofacial inflammatory pain model. Trigeminal activation

Trigeminal activation and sensitisation involved in chronic inflammatory orofacial pain share several similarities with headaches including migraine, which are still unmet medical need conditions. Therefore, understanding the pathophysiological mechanisms is important to determine novel therapies, in which validated animal models are necessary. Here we aimed to identify key mediators, mechanisms and networks in a rat orofacial inflammatory pain model. Experimental Approach: Complete Freund's Adjuvant (CFA, 50 µl, 1 mg/mL) was injected into the right whisker pad of male Wistar rats, mechanonociceptive threshold was measured by von Frey filaments. Transcriptomics of the trigeminal ganglia (TRG, n=11/group) was performed by next-generation sequencing, and analysed together with the plasma metabolomic data.
