Gain and loss of function genome wide CRISPR/Cas9 screens in LGSOC cell lines to identify genes that modulate response to MEK inhibitor trametinib LGSOC cells were transduced with genome-wide CRISPR knockout or overexpression libraries. After selection and recovery, a T0 sample was taken and transduced cells were plated in the absence (UT) and presence (TR) of trametinib. For resistance screens, cells were harvested after 3-4 weeks, for the enhancer screen after 10 days. Screen was performed in triplicate (I, II, III).
