Detect overall RNA level change under TET2 knockout in response to IFNγ. Loss of TET2 alters IFNγ transcriptome, TET2 mediates the IFNγ signaling pathway. The experiment is designed to determine the overall RNA level change between TET2 knockout and control group in response to IFNγ. Briefly, we first generated TET2 KO THP1 cell lines by CRISPR technology. Then we treated control and TET2 KO THP1 cells with 100ng/ml IFNγ for 20hrs, total RNAs from each group were extracted and sequenced, 3 replicates in each group and 2 technical repeat for each sample.
