Comparative analysis of gene expression differences (DEGs) between the amygdalohippocampal complex (most often the region of seizure onset in mesial temporal lobe epilepsy (mTLE)) and neocortex from mTLE patients who undergo epilepsy surgery can be used to illuminate pathophysiological events involved in epileptogenises. Transcriptome analysis was performed on freshly resected amygdalohippocampal and temporal lobe cortex tissue obtained after surgery of 17 patients with drug-resistant temporal lobe epilepsy from Copenhagen University Hospital. Hippocampal and neocortex samples from 17 mesial temporal lobe epilepsy patients (paired data), and neocortex from 2 patients without epilepsy results in a total of 36 samples. The tissue was surgically removed en bloc and cut to 50 mg pieces. After a short rinse in RNAlater tissue sample 1-13 was placed in Rnase-free eppendorf tubes and flash frozen in liquid nitrogen. Sample 14-17 was placed in RNAlater.
