Chloroplast phosphate translocators coordinate carbon allocation between the cytosol and plastid during heterotrophic growth. Here, RNA-seq was performed to characterize transcriptome reprogramming in Chlamydomonas reinhardtii following transfer from low light to extended darkness, comparing wild type (CC-4532 background) and an independent CreTPT10 knockout mutant (tpt10b). Overall design: Total RNA was collected from WT and tpt10b cultures at 0 h under low light (LL) before transfer, and after 24 h and 48 h in complete darkness. Three biological replicates were sequenced per condition using paired-end Illumina RNA-seq. Processed data include gene-level count and TPM expression matrices and differential-expression results for tpt10b versus WT at each timepoint.
