We placed immotile variants of the soil bacterium Pseudomonas fluorescens SBW25 (SBW25(delta)fleQ IS-(omega)Km-hah: PFLU2552) under selection for recovered flagella-mediated motility using soft agar as a directional selection medium. We observed that typically a single nucleotide polymorphism (SNP) would arise de novo within 72-120 hours and restore the motility phenotype. Throughout this experiment we identified a rare subset of mutant lines that acquired in-frame deletions in the ntrB locus, which reduced residues 136-140 (LVRGL) to a single L at residue 136 in the translated protein product. One of the identified mutations that caused this change (ntrB del.410-421) was sent for whole-genome sequencing to screen for secondary, background and hitchhiking mutations.
