The aim of this study was to evaluate the ability of DNA metabarcoding, by rbcl as barcode marker, to identify and classify the small traces of plant DNA isolated from raw milk used to produce one of the most popular Italian PDO (Protected Designation of Origin): Grana Padano (GP) cheese. A total of 42 GP bulk tank milk samples were collected from 14 dairies belonging to Grana Padano Protection Consortium and 4 commercial milk samples retrieved outside the GP production site. Moreover, one sample of Soy flour (Glycine max) and two feedstuffs allowed by GP PDO production specification rules were also collected as technical controls.
