The protective immune response produced by fish after vaccination is the core indicator of vaccine effectiveness. Our previous studies have shown that inactivated vaccine against Edwardsiella tarda (E. tarda) can induce an immune response in flounder. To elucidate the protective immune response at the genetic level in flounder, in this study RNA-Seq technique was applied to analyze spleens of inactivated vaccinated flounder (Paralichthys olivaceus) on the seventh day after immunization and on the first and seventh day after five weeks of immunization with E. tarda infection. The results showed that DEGs are involved in many immune processes, mainly Toll-like receptor signaling pathways, antigen processing and presentation, and T1 and Th2 cell differentiation. This study helps us to further understand the immune mechanism of protection by inactivated E. tarda vaccine in fish.
