High levels of transcription and replication increase DNA supercoiling to levels that can impede or halt these processes unless removed by DNA topoisomerases. As a potent transcription amplifier and accelerator of replication, the protooncogene MYC must contend with high levels of torsional stress. Whether or how MYC harnesses topoisomerases to manage DNA supercoiling has not been well-studied. To show colocalization of MYC and topoisomerases, especially at promoters of highly expressed genes, we performed high-resolution mapping of MYC and TOP2a occupancy in vivo by ChIP-seq and compared their distributions. This study provides relevant scientific findings for various specialist like molecular biologists and biochemists, as well as cancer biologists.
