As an important forage crop and turfgrass, white clover(Trifolium repens) has the characteristics of wide adaptability, high heat and cold resistance, good ecological and economic value, and is one of the main cultivated herbage in the world. Due to the lack of reference genome, breeding and cultivation of white clover artificial grassland face great challenges. Therefore, this study proposes a white clover chromosome level de novo genomic assembly.The latest PacBio third-generation HiFi assembly and sequencing revealed that the genome size of white clover was 1.09 Gb with GC content of 33.64% , contig N50=12.85Mbp, scaffold N50=64.54Mbp. Moreover HI-C assisted assembly reveal, a total of 1.04Gb genome sequences were located on 16 chromosomes, accounting for 95.06%. Further annotation analyses predicted 90,128 protein-coding genes , 46,089 transcripts and 15,520 non-coding RNA genes. Moreover On the basis of genome assembly sequence, the existing gene and genome sequence are compared to understand the genome structure, gene function and species evolution analysis.We demonstrated efficiency of the current state of the strategy for de novo assembly of the white clover, based on PacBio third-generation HiFi sequencing, high-throughput chromosome conformation capture technology, and PacBio SMRT technology. The proposed high-quality genome of White clover provides a key basis for accelerating the research and molecular breeding of this important forage crop.
