80 sequence samples were collected, involving 12 individuals. It contains two mitochondrial genes: cytochrome oxidase subunit I (CO ⅰ), cytochrome b (Cyt b), and five single copy nuclear genes: early growth response genes 1 and 2B (EGR1 and egr2b) Photoreceptor retinal binding protein (IRBP), recombination activating 2 (RAG2), rhodopsin 1 (Rh1). Take the muscle or fin of fish from each sample for DNA extraction and sequencing. Select the DNA extraction kit of Tiangen biochemical technology (Beijing) Co., Ltd. to process the collected muscle and fin tissue to obtain the whole genome DNA of the sample, and then use it for PCR amplification, All PCR products were tested and purified by 1% agarose gel electrophoresis and sequenced by Wuhan Ai Kangjian Biotechnology Co., Ltd. Double ended sequencing results were spliced using seqman (DNAStar, Madison, WI, USA).
