We detected numerous copy number aberrations in SNP arrays from follicular lymphoma patients. Additionally, we found an outstanding homozygous state involving folic acid-related genes. To study a hypothetical association between both traits, we cultured folate-depleted cell lines. After restoring normal medium to the cultures, we performed CGH arrays to examine the induced copy number aberrations. To study genome lesions in newly diagnosed follicular lymphoma patients, we performed 55 SNP arrays using DNA extracted from whole peripheral blood or bone marrow. 19 additional SNP arrays of DNA collected from patients during the follow-up were included in the homozygosity analysis. To evaluate the effects of folic acid depletion, we cultured five replicates of IM9 and DOHH2 cell lines during 15 days with 0 or 0.01 µM folate. Two culture replicates of cell lines with normal media (1 µM folate) were used as controls. After a 15-day folate deprivation, we restored normal media and cultured the cells for up to 1 month. CGH arrays were performed using DNA extracted from two IM9 samples and two DOHH2 samples. DNA pools of normal media culture replicates were used as reference samples.
