Low efficiency of targeted DNA integration and generation of undesired mutations represent major limitations for genome editing applications. To close this technology gap, we used modulators of DNA repair mechanisms and achieved extremely efficient and precise insertions of different DNA donor templates into eukaryotic genomes. Therefore CRISPR-Cas9 edited cells were subjected to Illumina deep targeted amplicon sequencing to analyse InDel and knock-in efficiencies.
