PacBio sequencing data of the edited PIGA gene locus (HCT116)

Sequencing was performed to quantify knock-in events as well as unintended insertions and deletions (indels) generated during the procedures of targeted knock-in. PacBio sequencing was employed to detect indels across an approximately 5.5 kb window. In practice, targeted knock-in of a mutation in PIGA exon 6 in the HCT116 cell line was carried out using tandem paired nicking and the conventional Cas9 nuclease-mediated knock-in approach. Genomic DNA was subsequently extracted from the bulk transfectants, and an approximately 5.5 kb region surrounding the knock-in site was PCR-amplified and subjected to PacBio sequencing.
