This dataset contains RNA sequencing (RNA-seq) results derived from human hepatocellular carcinoma cells (HepG2) cultured in a liver-on-a-chip microphysiological system. The organ-on-a-chip platform enables continuous perfusion and physiological shear stress, thereby better mimicking the in vivo hepatic microenvironment compared to conventional static culture systems. In this study, HepG2 cells were exposed to 10 uM of triphenyl phosphate (TPhP), an organophosphate flame retardant, for 48 hours under dynamic flow conditions.Total RNA was extracted after chemical exposure and subjected to high-throughput sequencing to capture transcriptomic responses associated with TPhP-induced hepatotoxicity. The liver-on-a-chip model provides improved metabolic competence and cell-to-cell interaction compared to traditional monolayer cultures, allowing for a more physiologically relevant assessment of chemical effects.
