The tumor microenvironment is a complex ecosystem that plays a critical role in tumor progression. In this study, we employed single-cell and spatial transcriptomics to construct an atlas of the entire murine colon during tumorigenesis under dietary influence. Through comprehensive analysis, we identified a distinct tumor cell transcriptomic profile in both single-cell and spatial datasets. By leveraging our own data as well as publicly available datasets, we have created a comprehensive single-cell resource that enables deconvolution of spatial transcriptomic datasets. Through deconvolution techniques and the utilization of algorithms to predict co-localization, we uncovered a tumor-specific circuit comprising IgA plasma cells, B cells, and neutrophils. Interestingly, these immune cell populations exhibited localization adjacent to tumors, suggesting their potential involvement in tumor expansion or size control. Our findings provide novel insights into the intricate cellular composition of the tumor microenvironment during murine colon tumorigenesis. Understanding the specific roles of IgA plasma cells, B cells, and neutrophils in tumor development could lead to targeted therapeutic strategies aimed at modulating their functions and ultimately improving cancer treatment outcomes.
