Transcription elongation factor S-II (TFIIS), also known as TCEA, plays a key role in stimulating RNAPII to bypass sites of oxidative damage or resume transcription following elongation arrest. Whereas correlations between transcription stress and genome instability have been established, the mechanisms underlying their impact on cell homeostasis and function remain poorly understood. We created the first Tcea1fl/fl mouse model. Homozygous inactivation of the Tcea1 gene in mice leads to embryonic lethality between E13.5 and 16.5. Tcea1fl/fl mice were intercrossed with CMV.Cre mice to isolate Mouse Embryonic Fibroblasts (MEFs) from E12.5 mice embryos. We propose to investigate if a transcriptional defect can lead to cellular malfunction and premature cell death or senescence, resulting in DNA damage-induced, accelerated aging and disease. KO3 and WT3 samples were not included in the analysis while WT1 and WT1b were merged at the fastq level.
