Leishmania (L.) infantum is the etiologic agent of visceral leishmaniasis (VL). In Brazil represents a serious public health problem. Studies have shown that regulation of immune response appears to depend on miRNAs. In canine VL due to cell immune suppression being determinant of disease progression, knowledge of miRNAs may be important for the pattern of change in immune response. Here, we suggest that post-transcriptional regulation, mediated by miRNAs, may play a role in immune response of dogs with VL. Fifteen dogs were used to form two groups for the study: 5 healthy dogs in Control group and 10 dogs naturally Infected with L. infantum in Infected group. Peripheral blood mononuclear cells (PBMC) isolated from total blood using gradient Histopaque 1077 (Sigma-Aldrich, St. Louis, MO, USA). After isolation, total RNA was extracted and 250 ng of total RNA of RIN (RNA integrity number) over than 8 was used to evaluate the miRNA profile using a microarray strip containing 291 canine specific probes. Raw intensity values were background corrected, log2 transformed and then quantile normalized by the software Expression Console (Affymetrix) using the Robust Multi-array Average (RMA) algorithm. Next, statistical analysis was performed in the Transcriptome Analysis Console software (Affymetrix) by ANOVA (fold change ± 2, p <0.05).
