RNA-Seq analysis to assess gene expression levels in MDA-MB-231 cells on treatment with 1-SO (2,8-diacetoxymethyldibenzothiophene-S-oxide) and 1-S (2,8-diacetoxymethyldibenzothiophene) under variable UV conditions We report the gene expression levels in MDA-MB-231 cells on treatment with 1-SO (2,8-diacetoxymethyldibenzothiophene-S-oxide) and 1-S (2,8-diacetoxymethyldibenzothiophene) under 30 minute UV-A irradiation (UV) and no UV-irradiation/dark (No-UV) Overall design: MDA-MB-231 cells (600,000 cells/1.8mL/well) were plated on a six-well plates. The plates were incubated with compounds (10uM-final concentration) for 24 hours (VC - 1% DMSO). For UV experiment, one plate was placed in a Luzchem photoreactor equipped with 8 LZC-UVA bulbs (fwhm, 325-375 nm) for 30 minutes and for the No_UV experiment, the plate was covered with an aluminum foil for 30 minutes. RNA was then extracted using a total purification RNA kit (Sigma Aldrich - Catalog #RNB100) based on the associated protocol. Novogene Corporation performed sequencing and associated bioinformatics analysis.
