Symbiodiniacese ITS2 data for "Stony Coral Symbioses Show Variable Responses to Future Ocean Conditions"

These datasets contain cleaned SymPortal outputs and associated experimental metadata for coral samples used to evaluate Symbiodiniaceae community responses after a long-term mesocosm experiment. Coral fragments were exposed to four experimental treatment conditions: Control, Acidified, Heated, and Acidified-Heated. The datasets also include Ambient Seawater samples, which represent the Symbiodiniaceae communities present in the original field-collected coral colonies at the time they were collected from their site of origin, prior to fragmentation and experimental exposure. These original colonies were later fragmented and used in the experiment. Therefore, Ambient Seawater samples do not have nubbin numbers, as nubbin identifiers were assigned only after fragmentation for the experimental treatments. Two SymPortal-derived datasets are included. Type_absolute.xlsx contains absolute abundance data for SymPortal-resolved Symbiodiniaceae types. Pro_absolute.xlsx contains absolute abundance data for SymPortal-resolved Symbiodiniaceae profiles. Many terms have been used to describe the Symbiodiniaceae unity of resolution. To avoid ambiguity, we restrict our use to ‘Symbiodiniaceae type’ and ‘Symbiodiniaceae profile’. A type refers to Symbiodiniaceae taxa that have a specific sequence as their most abundant sequence. A Symbiodiniaceae profile is a set of ITS2 sequences that have been found in a sufficient number of independent samples to be identified as a ‘defining intragenomic variant’ (DIV). For example, C17 is a Symbiodiniaceae type and C17d/C31-C21-C17e-C21ac-C17f-C17 g is a Symbiodiniaceae profile with C17d present in higher abundance than the other types. In the dataset, each row represents an individual coral fragment or original colony sample, with associated metadata including species, colony identity, site of origin, treatment, temperature condition, CO₂ condition, nubbin identifier, bleaching status and sample UID. These datasets were used for downstream analyses of Symbiodiniaceae community composition, diversity, and treatment-associated shifts across coral species, sites, and experimental stress treatments. Samples that did not yield sufficient high-quality DNA for downstream analysis were excluded from these cleaned datasets.
