To uncover additional gain-of-function properties of lactylation on ALDOA, we reasoned that this might result in regulated gene expression at transcriptional level based on previous findings on multiple metabolic enzymes. Driven by this speculation, we performed RNA-sequencing (RNA-seq) analysis on HEK293T cells that site-specifically encode ALDOA-K147 (147TAG+Klac) through co-transfection of KlaRS1/tRNACUA pair and ALDOA-147TAG plasmids and cultured with Klac, and used cells that were transfected in a similar manner but cultured without Klac as the control group (147TAG). Pairwise comparisons led us to identify 111 genes as differentially regulated (70 upregulated and 41 downregulated) by expression of ALDOA-147Klac.
