The skin has remarkable ability to grow under constant stretch. We developed a mouse model to imitate human tissue expansion to study the cellular and molecular mechanisms involved in tension-induced skin growth. We examined specific pathway changes within the entire skin at both the spatial and single-cell level. Our analysis has revealed the fractions of basal and suprabasal cells increased after expansion. We observed elevated infiltration of immune cells in the skin, suggesting a tension-induced skin inflammatory response. Expanded skin and nonexpanded skin of mice collected at time point PI-14 were used for analysis. Mice expanded skin with Yoda1 treatment and mice expanded skin without Yoda1 treatment collected at time point PI-7 were used for analysis. Samples from expanded tissue, nonexpanded control tissue, expanded tissue with Yoda1 treatment, and expanded tissue without Yoda1 treatment were analyzed using spatial transcriptomics.
