In this study, role of Mad2l2 in the regulation of embryonic stem cell's chromatin architect were studied with MeDIP-sequencing.5 methyl cytosine levels were increased in the absence of Mad2l2 generally with some exceptions on the promoter of maternal and paternal regulated genes. Super resolution microscopy showed Mad2l2 colocalization with open chromatin. Additionally, Mad2l2-deficient cells do not express Dppa3. Dppa3 is a DNA methylation regulator.
