Mycoplasma bovis (M.bovis) is a critical pathogen of bovines resulting in pneumonia, mastitis, arthritis, etc. To reveal its virulence related factors, a virulent M. bovis HB0801 and its derived vaccine strain P150 were infected calve and the transcriptome profiles of PBMCs were compared by using of the microarray at 7 days after infection. The data postulated the pathogenic mechanism of wild strain and immune mechanism of the attenuated strain to provide clues for the further research of the interaction between M. bovis and its host. Nine calve were nasal inoculated with 109 CFU of HB0801 strain or its P150 strain respectively,or correspond volume of PPLO broth as the negative control, with three calve in one group. After 7 days post-incubated, PBMCs were isolated from blood samples on a Ficoll gradient. Then the PBMCs were inoculated in a 12-well plate at a density of 2-4 × 106 live cells each well, and co-incubated with 2μg for each well heat-killed either HB0801 or P150 M. bovis strains. Transcriptome profiles of peripheral blood mononuclear cells (PBMC) were compared by using of the microarray.
