Transcriptional Profiling of Dorsal Root Ganglion Neurons Following Sciatic Nerve Injury

To elucidate transcriptional responses in peripherally injured neurons, we performed RNA-seq of dorsal root gangliona (DRG) sensory neurons at 1 and 5 days following sciatic nerve injury. Sciatic nerve crush was performed by exposing the left sciatic nerve at the level of sciatic notch and crushing with smooth forceps for 30s. L3-6 DRGs from naïve or crushed mice were dissected and vGLUT2-tdTomato labeled neurons were dissociated and selected by fluorescence-activated cell sorting (FACS), then mRNA purified and profiled by sequencing.
