Survival motor neuron (SMN) protein localizes ubiquitously and exerts it function in various manners. SMN also binds with RNA polymerase II (RNAPLII) via the C-terminal domain (CTD) for the transcriptional termination. Here we show the SMN transiently localizes in the nucleus during myogenic differentiation, and SMN physically interacts with Myoblast determination protein 1 (MYOD1). Additionally, SMN and MYOD1 colocalize in the promoter regions of some MYOD1-regulated genes. The depletion of SMN downregulates the expression of SMN-bound genes, suggesting the directly effect of SMN on transcriptional regulation in the promoter regions. Overall, our results suggest that SMN mediates the transcriptional regulation in promoter regions collaborating with MYOD1.Overall design: ChIP-seq with SMN, RNAPLII and FLAG-MYOD1 using iPSCs-derived MYOD1-driven myogenic cells at day 3 after myogenic conversion.
