Segmental duplications cause chromosomal variations in eukaryotic genomes. The process further changes the copy number of duplicated genes and leads to phenotypic changes that affect plant adaptability. Segmental duplications play roles in plant domestication and enrich genetic resources for plant breeding. The reduced height(Rht) locus Rht-D1c, which is a duplicated region of the Rht-D1b gene, confers an extreme dwarf phenotype in common wheat (Triticum aestivum). However, other than the status of segmental duplications, the sequence of duplicated Rht-D1b genes is unknown. In this study, bacterial artificial chromosomes (BACs) were constructedfrom a dwarf hexaploid wheat 'Lumai 15' that carries the Rht-D1c locus. BAC clones were identified using specific primers for Rht-D1b and were sequenced separately on a MiSeq platform.
