The enzyme was incubated at 37°C in 50 mM Hepes buffer pH 7.40 containing 1 mM EDTA and 5 mM DTT in the presence or absence of each modifier. Aliquots were withdrawn from the reaction mixtures at regular time intervals and their residual activity measured using the substrate Z-FR↓AMC.Effect of modifiers on the stability of cathepsin K at pH 7.4.
