The former silver mine "Reiche Zeche", situated in Freiberg, Saxony was founded in the late 14th century. Since 1919, the mine serves for research and educational purposes of the Technical University Bergakademie Freiberg. The high number of ore lodes in the mine is one of the reasons for the generation of acid mine drainage with partially high metal concentration. The current 16S rRNA-based metagenomic analysis of microbial communities composition from eleven habitats sampled in different wings of the mine aims at a deeper understanding for the occurrence and distribution of acidophilic microorganisms. Samples were taken from biofilm formations at gallery ceilings and side walls but also from stagnant and flowing acid mine drainage locations. Illumina sequencing technology (MiSeq) was applied for sequencing the V3V4 region of the 16S rRNA gene.
