The transcription factor ThPOK is critical for homeostasis and differentiation of mature T helper cells. Overall goal of this experiment is to show divergent gene expression profiles of ThPOK-dependent autoregulatory loop disrupted in CD4 T cells. Naïve T cells (TCRb+CD25-CD44- Cd62l hi) from WT (CD4 and CD8) and OB11/OB11 (CD4hi, CD4+CD8+, CD4lo) mice were sorted based on CD4 and CD8 expression. RNA was extracted using Trizol and were subjected RNA seq using illumina sequencing.
