Transcriptome profiling disclosed the effect of dark- induced leaf senescence (DILS) and developmental leaf senescence (DLS) on reprogramming of genes expression in barley first leaf of young seedlings (DILS) as well as in flag leaf of mature plants (DLS). Hordeum vulgare cv. Golden Promise seeds were sterilized and growth in soil for 7 days. For dark- induced leaf senescence 7 day- old seedlings were exposed to dark or left in growth light for 0 (control), 4, 7 or 10 days. First leaves of plants were collected (three leaves were combined into one replicate/sample). For developmental leaf senescence, flag leaves of mature plants were collected 5 days before anthesis (control) and 30 days after anthesis.
