Chromosome spreads were prepared from S. cerevisiae cells at 5.5 hr (pachytene) into meiosis. Plasmid foci were mapped with respect to Mps3 as the telomere marker. Chromosomes were visualized by DAPI (blue). Mps3 was visualized by an HA-directed antibody to Mps3-3HA and a Texas Red-conjugated secondary antibody (red). The plasmid was detected using an antibody to GFP (green), which targets the GFP-LacI bound to the LacO array present on the plasmid.
