Dendrobium nobile Lindley (DNL), a traditional Chinese medicinal herb, contains bioactive components, including Dendrobium nobile Lindley polysaccharides (DNLP) and Dendrobium nobile Lindley alkaloids (DNLA), which exhibit immunomodulatory and anti-inflammatory properties. Despite these promising characteristics, their therapeutic potential in psoriasis remains underexplored. This study investigated the effects of DNLP and DNLA in an imiquimod (IMQ)-induced psoriasis mouse model using techniques such as PASI scoring, histopathological analysis, immunofluorescence, RT-PCR, and RNA-Seq. Results demonstrated that oral administration of DNLP and DNLA significantly alleviated psoriasis symptoms and improved histopathological features. RNA-Seq analysis revealed that both components modulate immune responses and cellular processes, reducing inflammation by inhibiting myeloid leukocyte infiltration. Immunofluorescence further validated these findings by showing downregulation of keratin markers (K14, K16, K17) and reduced expression of macrophage and neutrophil markers in treated groups. The study concludes that DNLP and DNLA possess significant anti-inflammatory properties, acting through the regulation of keratinocyte differentiation and myeloid leukocyte activation. These findings highlight the therapeutic potential of DNLP and DNLA in psoriasis treatment.
