We developed an efficient, one-step method for constructing large CRISPR arrays. Three assembled CRISPR arrays of FnCas12a were subjected to small RNA sequencing after co-incubation with FnCas12a encoding plasmid in TXTL. The purpose of the NGS is to examine if the generated array can be processed correctly into individual crRNA.
