To characterize the spatial characteristics of the lung tissue of OP patient, we collected lung samples for Visium CytAssist from one patient with OP Overall design: The Visium spatial gene expression analysis was conducted using the Visium CytAssist Spatial Gene Expression for FFPE kits (10x Genomics, PN-1000520). The tissue slices underwent crosslinking treatment followed by probe hybridization and probe connection on the crosslinked sections. After connecting the probes, RNA from the tissue section was enzymatically digested using RNase with tissue removal enzyme (10x Genomics, PN-3000387) to release the probes from the cells. These released probes, which were captured by the capture sequences on the Visium CytAssist Spatial Gene Expression slide using the Visium CytAssist instrument, were then extended and eluted. Subsequently, the eluted probes were transferred to new tubes for further use.single-cell RNA-seq libraries were pooled and sequenced on a NovaSeq 6000 instrument.
