We cultured a parental cell line (M14) in the presence of increasing concentrations of BRAFi to select for BRAFi resistant mutants. We then performed RNAseq on the parental cell line and the derived BRAFi resistant mutant cell line to elucidate the mechanisms of acquired BRAFi resistance.
