The circadian gene expression in peripheral tissue displays rhythmicity which is reprogrammed by feeding rhythms and dietary composition in mammals. In this study, circadian transcriptome was performed to investigate how the circadian clock and mineralocorticoid receptor contribute to daily gene expression in the colon. Overall design: 8- to 12-week-old, gut-specific Bmal1 knockout mice and MR knockout mice were subjected to ad libitum normal chow diet. 8-week-old male C57BL/6 mice were subjected to time-restricted feeding of low- or high-salt diet under 12hr light/ 12hr dark schedule. Tissues were harvested every 4 hours over the circadian cycle at ZT0, 4, 8, 12, 16, 20 (n=3 per time point per group). Total RNA was extracted from the colon, and used for RNA-seq.
