To investigate the effect of RNA-binding motifs of heterogeneous nuclear ribonucleoprotein K on activation-induced cytidine deaminase-dependent somatic hypermutation, NGS was performed using a pooled DNA library developed from 10 samples. For each sample, a 447 bp region targeting the 5'-Smu of immunoglobulin heavy chain gene was amplified and followed by index PCR using the Nextera XT Index kit (Illumina, FC-131-1001) according to the manufacturer's protocol (16S Metagenomic Sequencing Library Preparation, 15044223 B JPN, Illumina). The pooled library was sequenced on the Illumina MiSeq platform (2 x 300 bp).
