Argonaute (Ago) proteins are widely expressed in almost all organisms. Eukaryotic Ago (eAgo) proteins play a critical role in the biogenesis of small RNAs and prokaryotic Ago (pAgo) proteins defend against invading nucleic acids via binding small RNA or DNA. Protein domains are fundamental units of protein structure, function, and evolution, whereas the domains of Ago proteins are not well annotated/curated currently. Therefore, a comprehensive domain annotation database of Ago proteins is lacking, and it is not conducive for researchers to understand the function and mechanism of argonaute proteins. Herein, we constructed the first online resource (AGODB), a comprehensive domain annotation database of Ago proteins. AGODB curates domain annotation, specially for long pAgos, all six domains are annotated/curated. The database also allows users to search the database with a variety of options and download the search results.
Argonaute(Ago)蛋白几乎在所有生物中均广泛表达。其中,真核Ago(eAgo)蛋白在小RNA的生物发生过程中发挥关键作用,而原核Ago(pAgo)蛋白则通过结合小RNA或DNA来抵御入侵的核酸。蛋白质结构域是蛋白质结构、功能与进化的基本单元,但目前Ago蛋白的结构域尚未得到充分注释与整理。因此,当前缺乏一套完整的Ago蛋白结构域注释数据库,这不利于研究人员深入理解Argonaute蛋白的功能与作用机制。在此,我们构建了首个Ago蛋白结构域注释综合在线数据库(AGODB)。AGODB对Ago蛋白的结构域进行了系统性注释,其中针对长型原核Ago蛋白,已完成全部6个结构域的注释与整理。该数据库支持用户通过多种检索条件进行查询,并可下载检索结果。
