To determine the mechanism of regulation of Tregs by Setd2, SPTregs (CD3+CD4+Foxp3-YFP+) from Setd2-deficient and control Tregs were subjected to RNA-seq analysis. About 1 × 10^6 sorted splenic Tregs (CD3+CD4+YFP+ cells) were pooled from littermate Foxp3Cre-YFPSetd2f/+ or Foxp3Cre-YFPSetd2f/f mice. Each biological replicate was pooled from 2 mice.
