The goal of this study is to find out the relationship between ER-mediated transcription and DNA damage induced by estrogen(E2) and BRCA1 deficiency in MCF7. The DNA damage is marked by chromatin binding of γH2AX using ChIP-seq. We used ChIP-seq to obtain chromatin binding of γH2AX and ER in BRCA1 knockdown and control MCF7 with or without 10nM E2 treatment for 12h. DRB is used to inhibit transcription. mRNA profiles were generated by RNA-seq in same condition.
