We investigated gene expression changes of glucose deprived MCF-7 and T47D breast cancer cells supplemented with either 10 mM or 25 mM BHB. Glucose deprivation revealed numerous differentially expressed genes indicating an involvement of the Hippo pathway in MCF-7 cells and the NRF2-Ferroptosis axis in T47D cells. Beta-hydroxybutyrate had limited impact on breast cancer cells and differentially expressed genes were not associated with any pathways following pathway enrichment analysis. Two established human breast cancer cell lines were used for this analysis, MCF-7 and T47D. Cells were treated in either 225 mg/l glucose (5%) or 4.5 g/l glucose (100%). Cells treated with 225 mg/l glucose were additionally treated with no, 10 mM or 25 mM beta-hydroxybutyrate. Each condition was analyzed in duplicate except the comination glucose5%_BHB10mM in MCF-7 cells since one of the replicates failed library construction.
