Acquisition of cross-azole tolerance and aneuploidy in Candida albicans strains evolved to posaconazole

We sought to determine the diversity of phenotypic and genotypic changes that occur when the human fungal pathogen Candida albicans is evolved to the antifungal drug posaconazole. In the lab, we evolved replicates of the C. albicans strain SC5314 to YPD + 0.5 ug/mL posaconazole for 50 generations using batch culture evolution (1:1000 dilutions every 72 h). At the end of the experiment, we whole genome sequenced two ancestral SC5314 replicates and twelve evolved replicates.
