RNA seq analysis of MC38CEA cells with diminished ADAM17 expression due to Crispr/Cas9 gene editing or control cells

The goal of this study was to compare transcriptomes of cells with reduced and normal levels of ADAM17, unstimulated or stimulated with IL1b and IFNg Overall design: The cells were electroporated with Cas9 mRNA and after four hours electroporated with in vitro transcribed sgRNAs targeting Adam17 or EGFP (control). The cells were left unstimulated or were stimulated with human recombinant (hr) IL1b (10 ng/ml) and mouse recombinant (mr) IFNg (10 ng/ml) for six hours and RNA was isolated.
