Ex vivo cultured human cord blood CD34+ cells were sorted as Mitochondrial ROS low cells(15%) and Mitochondiral ROS high cells (15%). These two populations were analyzed by single cell RNA-seq. Totally 18 cell clusters were identified by UMAP analysis.One of the clusters enriched with AVP, HLF,CD34, PROM1, MLLT3 and MSI2 was proved by in vivo transplantation experiments as the functional hematopoietic stem cell population. ADGRG1 combined with CD34 and PROM1 could be used to enrich functional hematopoietic stem cell upon ex vivo culture stress. Fresh humn cord blood CD34+ cells were ex vivo cultured in Stemline II medium with growth factors. At day 4, cells were sorted by FACS as Mitochondiral ROS low and Mitochondrial ROS high CD34+ cell populations. 10X single cell RNA-seq analysis was performed to analyze these two CD34+ cell populations with different mitochondrial ROS levels.
