DNA polymerase ? synthesizes both strands during break-induced replication

We applied our ribonucleotide mapping technique HydEn-seq to study DNA polymerase usage during break-induced replication (BIR) in the budding yeast Saccharomyces cerevisiae. Overall design: A genetic assay of BIR was designed to induce a single break on the left arm chromosome X which triggers recombination with homology donor on Chromosome VI and leads to BIR. The BIR product (Chromosome VI/X hybrid) was isolated in polymerase mutants promiscuous for ribonucleotide incorporation. HydEn-seq was performed to map ribonucleotides in the BIR products. Data was anylzed to discern polymerase usage during BIR. Please note that the BIR_* sample processed data was generated from both replicates (BIR_*_1 and BIR_*_2 samples) and is linked to the corresponding BIR_*_1 sample records.
